I still think Spaceballs belongs at the top of the list. I've seen it a million times and it still makes me laugh so hard...often before the joke even comes.

Uncle Buck -- has to be John Candy's best roll.
Tommy Boy -- another movie you can watch over and over again
Revenge of the Nerds -- Classic Comedy, if there is such a genre
Groundhog Day -- another Bill Murry classic (Caddyshack rates higher though)
The Birdcage -- Nathan Lane is hilarious.
